Physicochemical and structural properties of soluble jumbo squid (Dosidicus gigas) elastin recovered from skin by-products were evaluated. The molecular weight of isolated elastin was ~40 kDa with an isoelectric point (pI) between 9 and 10. Aspartic, glutamic, arginine, proline, glycine, and lysine amino acids were the most abundant in squid elastin, whereas the hydroxyproline absence, ~0.7% cysteine content, and the calculated 0.35 isoleucine/leucine ratio were used as purity index. Total and reactive sulfhydryl contents were similar (247.0 ± 5.1 vs 242.0 ± 7.5 μmol mg?1 of protein, P ≥ 0.05) in purified squid elastin but surprisingly higher than previously reported in other elastins. On the other hand, the secondary structures of squid elastin analyzed by Fourier transform infrared spectroscopy (FTIR) were ~45% β-sheets, ~15% α-helices, ~10% β-turns, and ~30% undefined structures. In addition, squid elastin experienced glass transition at 82.01 ± 0.01ºC, denaturation temperature at 110.45 ± 0.64ºC, and aggregation at 197.5 ± 0.23ºC. In conclusion, the prevalence of charged amino acids and pI of squid elastin can facilitate its solubilization in hydrophilic systems, whereas the secondary structure profile and thermostability are desirable features in proteins used for biopolymer designs such as food biofilms or barrier systems. 相似文献

适度卷曲有利于提高水稻叶片的光合效率,增加植株光合产物的有效积累量。我们利用甲基磺酸乙酯(EMS)处理籼型水稻保持系西农1B,获得一个稳定遗传的水稻半外卷叶突变体。该突变体从十叶期开始各叶片逐渐向外卷曲直至半卷状,并伴随茎秆半矮化和叶片披垂,暂被命名为semi-outcurved leaf 1(sol1)。与野生型(WT)相比,sol1的叶片卷曲指数均达到30%以上(P<0.01);倒一、倒二、倒三、倒四节节间长度和穗长极显著缩短,倒一、倒二、倒三叶的叶夹角显著或极显著增加;有效穗数、千粒重、每穗实粒数、结实率显著或极显著下降,一次枝梗数则增加11.3%(P<0.05)。sol1的蒸腾速率、胞间CO2浓度、气孔导度显著高于野生型。石蜡切片显示,sol1倒一叶的泡状细胞体积变小,数量显著增多,表皮细胞体积略微增大。遗传分析表明,sol1的半外卷叶性状受1对隐性核基因调控,定位于6号染色体标记JY6-3和JY6-10之间165 kb的物理范围内,共含15个注释基因。qRT-PCR结果表明,与泡状细胞相关的内卷基因和外卷叶基因RL14、Roc5、REL1在突变体sol1中呈不同程度的上调,NRL、BRD1、OsHox32、ADL1、LC2则呈不同程度的下调。研究结果为SOL1基因的克隆和功能研究奠定了基础。 相似文献

对原核表达的重组建鲤组织蛋白酶L(Cathepsin L,CAT L)蛋白进行尿素洗涤和Ni-NTA亲和层析纯化,该目的蛋白经300 mmol/L咪唑洗脱为单一峰,SDS-PAGE结合TSK-GEL G2000SWxl凝胶过滤高效液相色谱分析表明重组CAT L获得了高度纯化,分子量约28 k D,纯度超过95%。Z-Phe-Arg-MCA底物测活法显示该重组CAT L表现为半胱氨酸蛋白酶活性,能与其内源抑制因子Cystatin以1︰1的摩尔比结合,具有生物学活性。以纯化的重组CAT L蛋白免疫Balb/C小鼠获得抗血清,经ELISA法检测获得的CAT L抗血清效价高于1︰512000;Western blotting鉴定结果表明该抗体具有良好的特异性,能够识别原核表达的重组CAT L蛋白。免疫组织化学分析结果表明,该抗体还能识别建鲤小肠、肝胰脏、脾、背肌和心肌组织表达的内源性CAT L蛋白。因此可利用该抗体从蛋白水平检测CAT L在鱼类不同组织中的表达和分布情况。 相似文献

以竹炭粉、陶土为主要原料,经混合、成型、干燥和煅烧等工艺制得竹炭陶土复合材料(以下称竹炭陶),采用扫描电子显微镜(SEM)、X射线衍射(XRD)仪、拉曼光谱测试仪(Raman)、傅里叶变换红外光谱(FTIR)和比表面积测试仪等仪器,对竹炭陶的微晶构造、孔隙结构、吸附和红外辐射性能等进行表征。结果表明:竹炭微粒镶嵌在陶土基体中,保留原有以中孔为主的孔隙结构和类石墨化晶体结构,获得的竹炭陶具有较大的比表面积和中孔为主的孔隙结构,能有效地吸附甲醛、苯等有害气体。加入一定量的竹炭能提高竹炭陶的远红外发射率,其红外发射率均高于陶土和竹炭,结合红外吸收光谱谱图分析,竹炭陶具有高红外发射率的本质在于具有倍频吸收、分子基团振动、转动及晶格振动。 相似文献

Amplified fragment length polymorphism (AFLP) analysis is a rapid and efficient method for producing DNA fingerprints and
molecular characterization. Our objectives were to: estimate genetic similarities (GS), marker indices, and polymorphic information
contents (PICs) for AFLP markers in almond cultivars; assess the genetic diversity of almond cultivars and wild species, using
GS estimated from AFLP fingerprints and molecular characterization; and facilitate the use of markers in inter-specific introgression
and cultivar improvement. The genetic diversity of 45 almond cultivars from Iran, Europe, and America, were studied assaying
19 primer combinations. In addition, several agronomic traits were evaluated, including flowering and maturity times, self-incompatibility,
and kernel and fruit properties. Out of the 813 polymerase chain reaction fragments that were scored, 781 (96.23%) were polymorphic.
GS ranged from 0.5 to 0.96, marker indices ranged from 51.37 to 78.79, and PICs ranged from 0.56 to 0.86. Results allowed
the unique molecular identification of all assayed genotypes. However, the correlation between genetic similarity clustering
as based on AFLP and clustering for agronomic traits was low. Cluster analysis based on AFLP data clearly differentiated the
genotypes and wild species according to their origin and pedigree, whereas, cluster analysis based on agronomic data differentiated
according the pomological characterization. Our results showed the great genetic diversity of the almond cultivars and their

本文概述了大豆营养特性与施肥的研究结果,提出了高产施肥的关键技术。大豆营养特性:一是对主要营养元素的吸收量较稻、麦作物高(等量籽粒产量),生产100kg大豆约吸收6.5~8.5kg N,1.8~2.8kg P2O5,2.7~3.7kg K2O,3.5~4.8kg CaO,1.8~2.9kg MgO,4.5~9.5g Zn。二是对主要营养元素的吸收积累高峰在花荚期,N、P、K的60~70%在在此期吸收,而不同于稻麦等作物。三是总氮源的40~60%来源共生固氮,而共生固氮又受土壤N、P、K、Ca、Mo、Zn等及土壤PH值影响。四是大豆成熟阶段营养器官的养分向籽粒转移率高,N、P、K分别达58~77%,60~75%,45~75%。大豆施用N、P、K、Zn、Mo、B肥均显著提提高产量,合理配合施用可达到180~300kg/亩。N、P提高籽粒蛋白质含量,K与Zn提高脂肪含量,P、K、Zn、Mo及少量N肥可提高结瘤固氮率。N、P、K和多种微肥可减轻东北连作大豆的不利因子危害,大幅度增加产量。大豆高产施肥,一是根据土壤、植株养分含量确定施肥数量,二是有机肥与N、P、K肥及多种微肥配合施用,三是注重前茬作物施肥,增肥土壤,四是根据土壤条件、耕作制度、大豆品种特性确定施肥数量、方法、时期。中等肥力高产施肥一般应施用N 8,P2O5 4,K2O 4,ZnSo4 1.5(kg/亩),钼酸铵20~30g/亩,P、K、Zn用作底或种肥,N肥钼肥种肥花期追肥各半,另于花荚期喷施P、N、Mo肥二次 相似文献

Summary Meiosis was studied in 31 wild Saccharum relatives, including Erianthus (8 clones), Miscanthus (5 clones), Narenga prophyrocoma (1 clone), S. robustum (3 clones), and S. spontaneum (14 clones). Chromosome number for 18 clones confirmed published counts or was typical of the particular species. Chromosome number for seven clones (Djatiroto 2n=58, Molokai 5099 2n=80, SES 84/58 2n=58, SES 114 2n=64, SES 260 2n=64, Taiwan 100 2n=112, and US 57-11-2 2n=60) differed from published counts (2n=112, 86-100, 64, 60, 60, 96, and 30, respectively). Counts were obtained for the first time for six clones (Local escape 2n=96, Nepal 2n=72, NG 77-77 2n=108–112, NG 77-199 2n=166, US 57-60-2 2n=20, and US 68-1-4 2n=38). Bivalent chromosome pairing predominated in all clones. Meiotic irregularity (numeric aberrations, univalents, multivalents, and telophase II micronuclei) tended to be associated with taxonomic grouping and level of polyploidy. Clones in Erianthus, Miscanthus, and Narenga were apparent euploids (2n=20–60) and tended to have fewer meiotic irregularities than Saccharum clones. Differences in level of meiotic stability among taxonomic groups may reflect error in chromosome association and synapsis associated with high chromosome number. 相似文献
